Mechanosensing of latrophilins: how GAIN domain translates force into function

Adhesion GPCRs sense mechanical cues for proper cellular functioning, but its activation mechanism remains unclear. This project aims to interrogate the contributions of the conserved GAIN domain, proposed to be a force-function translator, towards adhesion GPCR mechanosensing, using ADGRL3 as a model receptor. By developing a novel force application system, I will monitor structural alterations within the GAIN domain by force, and how its tertiary structure affects receptor mechanosensitivity.
